L'azienda americana ha annunciato oggi di aver presentato la domanda per ottenere dall'Agenzia europea del farmaco Ema la variazione dell'autorizzazione condizionata all'immissione in commercio. La valutazione è per un regime di 2 dosi da 50 microgrammi (metà rispetto al dosaggio destinato alle fasce d'età successive) del vaccino mRna-12731 nei 6-11enni.
"Siamo lieti di annunciare la presentazione di questa variazione all'Ema per l'uso del nostro vaccino Covid nei bambini di età compresa tra 6 e 11 anni nell'Unione Europea. Questo segna la nostra prima richiesta per l'uso del vaccino in questa fascia d'età", afferma Stéphane Bancel, Ceo di Moderna. "Siamo incoraggiati dal fatto che il vaccino Moderna al livello di dose di 50 µg abbia aiutato a prevenire l'infezione da Sars-CoV-2 nei bambini - ricorda - Abbiamo in programma di inviare questi dati ad altre agenzie di regolamentazione in tutto il mondo per proteggere questa importante popolazione più giovane".
Lo studio di fase 2 'KidCove' sull'mRna-1273 è in corso nella popolazione pediatrica di età compresa tra 6 mesi e meno di 12 anni. E Moderna ha recentemente condiviso nuovi dati per la fascia 6-11 anni. E' stata osservata un'efficacia del vaccino del 100% (utilizzando la definizione del caso primario P301 per Covid-19) 2 settimane dopo la prima dose di mRna-1273, ricorda l'azienda. Inoltre, per l'infezione asintomatica l'efficacia del vaccino 2 settimane dopo la prima dose era del 65%; per l'infezione da Sars-CoV-2 indipendentemente dai sintomi è stata dell'80%.
Il 24 ottobre la società ha annunciato anche dati positivi di prima linea dallo studio di fase 2/3 sul vaccino mRna-1273 in bambini tra 6 e meno di 12 anni: il rapporto delle medie geometriche (Gmr) dei titoli di anticorpi neutralizzanti anti Sars-CoV-2, che confrontava la risposta nei bambini con la risposta nei giovani adulti dello studio Cove di fase 3, era 1,5, con un tasso di sierorisposta del 99,3%. Le due dosi da 50 µg sono risultate generalmente ben tollerate.
Lo studio KidCove è uno studio di espansione randomizzato, in cieco per l'osservatore, controllato con placebo per valutare la sicurezza, la tollerabilità, la reattogenicità e l'efficacia delle due dosi somministrate a bambini sani a distanza di 28 giorni. La popolazione in studio è suddivisa in 3 gruppi di età (da 6 a meno di 12 anni, da 2 a meno di 6 anni e da 6 mesi a meno di 2 anni).
